 Action :

Anonsteroidal potent antioestrogen. It competes with oestradiol for oestrogen receptor protein in target tissues. Actions are similar to clomiphene.

  

 Indication :

Palliative treatment of advanced or metastatic carcinoma of breast.

  

 Pharmaco-Kinetics :

It is given orally, the drug has biphasic half life.

  

 Contraindication :

Hypersensitvity.

  

 Adverse-Effects :

Nausea, vomiting, hot flushes, vaginal bleeding, dermatitis, pruritus vulvae, menstrual irregularities.

  

 Special Precaution :

Premenopausal women.

  

 Interactions :

Oral anticoagulants

:

Significant increase in anticoagulant effect requiring careful monitoring of prothrombin time.

Bromocriptine

:

Serum tamoxifen levels are elevated.

Cyclophosphamide

:

Since tamoxifen is a potent inhibitor of cytochrome P450 oxidase it may affect metabolism and excretion of cyclophosphamide.

  

 Dosage :

10-20mg twice daily (morning & evening).

  

 Duration of action :

Terminal half-life > 24 hours.

  

 Strength & Packing :

10mg tablet, 
20mg tablet,  10 tablets per box.
