Kanamycin  -  Aminoglycoside antimicrobial

  

 Action :

Bactericidal by inhibiting synthesis of bacterial proteins. Susceptible organisms include staph. aureus (including penicillinase-producing strains), Staph. epidermidis, N. gonorrhoeae, H. influenzae, E. coli, Enterobacter, Shigella, Salmonella, Klebsiella pneumoniae, Serratia, Providencia, and many strains of Proteus. Less active against Streptococci and enterococci.

  

 Indication :

Staphylococcal, genito-urinary & respiratory infections. Infections resistant to other antibiotics. Drug resistant tuberculosis.

 

 Pharmaco-Kinetics :

It is given by parenteral route.

   

 Contraindication :

Hypersensitivity, severe renal failure.

  

 Adverse-Effects :

Nephrotoxicity, ototoxicity, neuromuscular blockade, headache, skin rash, urticaria, nausea, vomiting.

  

 Special Precaution :

Impaired renalfuction, myasthenia gravis.

  

 Interactions :

Amphotericin B, Cisplatin, Cephalosporin, Vancomycin, Methoxyflurane, Frusemide, Bumetanide

:

Increasednephrotoxicity.

Anaesthetics, Neuromuscular blocking agents (Tubocurarine)

:

Risk of neuromuscular blockade and respiratory paralysis.

Pencillin, Cephalosprins, Carbenicillin, Ticarcillin

:

Exert antibiotic synergism, especially useful in Pseudomonas infections.

Loop diuretics

:

Concurrent use of these agents may increase the risk of respiratory paralysis and renal dysfunction.

Lab Tests : Aminoglycoside serum levels

:

Guard against in vitro inactivation of aminoglycosides by B-lactum antibiotics in patients on combination therapy.

   

 Dosage :

Adults : 1g. daily. 

  

 Duration of action :

Half-life about 3 hours.
