Atracurium Besylate  -  Neuromuscular Blocker

  

 Action :

It is a competitive antagonist of acetylcholine at neuromuscular junction and cause flaccid paralysis of skeletal muscles. The neuromuscular block of this type can be reversed by administration of anticholinesterase drug (neostigmine) which inhibits the enzyme cholinesterase so that acetylcholine is not degraded. Hence the concentration of acetylcholine increases which tends to decrease the effect of neuromuscular blocker.

  

 Indication :

For muscle relaxation in surgical anaesthesia.

 

 Pharmaco-Kinetics :

Atracurium is unique in that it is altered in body to an inactive form by passive chemical process (Hofman elimination). Duration of action is not influenced by the state of circulation, the liver or kidneys, a real advantage in patients with hepatic or renal disease and in elderly.

   

 Contraindication :

Hypersensitivity.

  

 Adverse-Effects :

Hypotension, flushing, bronchospasm.

  

 Special Precaution :

Severe cardiovascular disease, severe electrolyte imbalance, myasthenia gravis, long term infusion. It should be administered with adeqyate general anaesthesia.

     

 Dosage :

Induction :  0.3 - 0.6mg / kg body weight I.V.

Maintenance :  0.1 - 0.2mg / kg body weight I.V.

       

 Duration of action :

20 - 40 minutes. 

    

 Strength & Packing :

10mg / ml,  5ml  vial,  Injection.
