Vincristine  Sulphate   -   Antineoplastic

  
Manufacturers of  Vincristine Sulphate injection, an anti neoplastic drug, used in the treatment of acute leukaemias, wilms tumor

  

 Action :

Vincristine arrests cell division at the metaphase stage by inhibiting microtubule formation in the mitotic spindle.

  

 Indication :

Acute leukaemias, lymphomas & certain other malignant conditions, neuroblastoma, wilms tumor other : idiopathic thrombocytopenic purpura.

  

 Pharmaco-Kinetics :

After oral administration the absorption is unpredictable. It is metabolised in liver.

  

 Contraindication :

Intrathecal administration, demyelinating charcot marie tooth syndrome.

  

 Adverse-Effects :

Local reaction if extravasation occurs, constipation, paralytic ileus, jaw pain, alopecia, bone marrow depression, peripheral neuropathy. Inappropriate ADH secretion. Shortness of breath, bronchospasm, hypo or hypertension.

  

 Special Precaution :

Infection, neuromuscular disease, leucopenia, pulmonary disorders, leukaemia, radiation therapy, extravasation, concomitant neurotoxic drugs. Eye contact, concurrent vaccination.

  

 Interactions :

Digoxin

:

Decreased plasma digoxin levels and renal excretion.

L-asparginase

:

Hepatic clearance of vincristine is reduced if L-Asparginase is administered first. Thus to minimize toxicity, vincristine should be given 12 to 24 hours before administering L-asparginase.

Mitomycin C

:

Acute pulmonary reaction may occur.

Phenytoin

:

Reduced phenytoin plasma levels.

  

 Dosage :

Adults

:

1.4mg / sq. m. body surface / weekly.

Children

:

   2mg / sq. m. body surface / weekly.

  

 Onset of action :

15 - 30 mins. following I.V. administration.

  

 Duration of action :

19 - 155 hours.

  

 Strength & Packing :

1mg Injection, 
5mg Injection,  single vial per box.
