Acute leukaemias, especially acute myelogenous leukaemia & acute lymphoblastic leukaemia. It is also used in the treatment of chronic granulocytic leukaemia.
Pregnancy, lactation, prior resistance to drug.
Myelosupression, cytopenia, hyperuricaemia, nausea, vomiting, anorexia & stomatitis.
Myelosuppression is preduced by thioguanine. Evaluation of haemoglobin concentration or haematocrit, total white blood cell count, differential count and platelet count is necessary during the therapy. When used in combination with other drugs whose primary toxicity is myelosuppression, life-threatening infections and bleeding have been observed as a consequences of thioguanine induced granulocytopenia and thrombocytopenia.
Complete cross-resistance between mercaptopurine and thioguanine. Exercise caution during long-term continuous therapy with busulfan.
Exact
dose and duration of administration will depend on the nature and dosage
of other cytotoxic drugs given in conjunction. Induction therapy :
100-200mg / sq. m. body surface area / day on a single or twice daily
dosing regimen over a period of 5-20 days.
40mg tablet, 25 tablets per box.
