Mercaptopurine  -   Antileukemia

  
Manufacturers of Mercaptopurine tablets, an anti leukaemia drug, used in combination therapy for acute leukaemia

  

 Action :

Inhibits nucleic acid biosynthesis and is active against leukaemias.

  

 Indication :

In combination therapy schedules for acute leukaemia, choriocarcinoma, chronic granulocytic leukaemia.

    

 Pharmaco-Kinetics :

After oral administration absorption is incomplete and the drug is metabolised in liver. The active metabolites have longer half life than parent drug.

  

 Adverse-Effects :

Nausea, vomiting, diarrhoea, cholestasis, bone marrow depression, pancreatitis, oral and intestinal ulcers. rarely hepatic necrosis.

  

 Special Precaution :

Impaired renal or hepatic function, monitor uric acid levels, blood counts and hepatic function.

  

 Interactions :

Allopurinol

:

Delayed catabolism of mercaptopurine resulting in severe toxicity. Dose of mercaptopurine should be reduced to one third to one quarter of usual dose.
Other myelosuppressive agents : Enhance anti-neoplastic effect of mercaptopurine, dose of mercaptopurine may need to be reduced.

Trimethoprim- sulfamethoxazole

:

May result in enhanced bone marrow suppression.

  

 Dosage :

Adults & Children : Usually 2.5mg / kg body weight daily. 

      

 Strength & Packing :

50mg tablet,  25 tablets per box.
