Enters body folate pool as 5-methyltrahydro folate. Can neutarlise the effects of folic acid antagonists such as methotrexate but increases those of 5-FU.
Over dosage of methotrexate, folic acid antagonists, adjuvant treatment of colorectal carcinoma.
Well absorbed by oral or intramuscular administration and is rapidly converted to biologically active folates distribution occurs to all body tissues and it is concentrated in the CSF. It is excreted in the urine.
Pernicious anaemia and megaloblastic anaemia secondary to the lack of Vit. B12.
Rarelypyrexia.
Should not be administered with folic acid antagonists.
As an antidote to methotrexate (started 8-24 hours after the beginning of methotrexate infusion) in general up to 120mg in divided doses over 12-24 hours by I.M. or I.V. injection or infusion followed by 12-15mg I.M. every 6 hours for the next 48-72 hours. Suspected methotrexate over doses, immediate administration of in a dose equal to the dose of methotrexate.
Terminal elimination half-life 6 hours.
