The predominant effect of etoposide is inhibition of DNA synthesis resulting in arrest of cell cycle. At low doses etoposide inhibits cells from entering cell cycle. At high doses cells entering mitosis are lysed.
Small cell lung cancer. Acute monocytic and myelomonocytic leukaemias. Hodgkin's disease. Non-Hodgkins lymphoma. testicular tumours.
After oral administration about 50% drug is absorbed.
History of previous hypersensitivity. Severe bone marrow failure not due to malignant disease, severe hepatic dysfunction.
Nausea, vomiting, diarrhoea, allergic reactions, fever, hypotension, bone marrow depression, peripheral neuropathy. Jaundice, bronchospasm.
Extravasation, infection, children, adequate contraception. Monitor hepatic, renal and haematological function.
Warfarin
: Prolongation of prothrombin time may occur.
Terminal half-life 3-19 hours.
