It forms a stable complex with DNA and interferes with the synthesis of nucleic acids. It is a cell-cycle nonspecific agent, but its cytotoxic effects are most marked on cells in the S-phase. It also has immunosuppressant action, and in addition has antibacterial effect.
Acute lymphocytic and acute granulocytic leukaemias. Acute myeloblastic leukaemia. Rhabdomyosarcoma and disseminated neuroblastoma.
Hypersensitivity to Daunorubicin or any component, CHF or arrhythmias, preexisting bone marrow suppression. Previous full cumulative doses of doxorubicin and / or extravasation.
Nausea, vomiting, bone marrow depression, stomatitis, alopecia, rash, hyperpigmentation, cardiotoxicity, CHF, fever and chills. Diarrhoea, severe local tissue damage and necrosis on extravasation. Anaphylactoid reaction, red urine (not haematuria).
Cardiac, haematological monitoring. Hepatic, renal impairment, children, extravasation.
Immunisation with live vaccines not recommended as daunorubicin is immunosuppressant.
Reconstitute vial with 4ml water for injection to make 5mg/ml solution. Dilute requisite dose with infusion fluid to a concentration of 1mg/ml and infuse over 20 mins.
Elimination half-life of daunorubicin 18.6 hours. Its metabolite 26.7 hours.
